The Hanging Stars Over The Silvery Lake Reviewed by Nathan Ford Ive been following the progress of Londons Hanging Stars with great interest via their first two 7"s (reviewed here and here), and am pleased to report that their first full length (also released by wonderful Indie label The Great Pop Supplement) more than lives up to the promise of those singles. And for those who missed those long out of print singles, their three best tracks are reprised here. This review is rather late in the game as "Over The Silvery Lake" has been out for a few months now, so apologies to both the band, and you dear reader, but if I may rely on a somewhat tired cliche, some things are worth waiting for, and "Over The Silvery Lake" is certainly getting better with age. The developmental proteome of Drosophila over 15 life stages! Drosophila melanogaster is the geneticists tool! What is someone doing stealing it and doing protein work on it?!?  Oh, you know, just finding out more about it than we ever knew! You can check it out in this new paper in GENOME RESEARCH (yeah!) here! They sampled fruit flies at 15 stages of their development -- including early stages that have nauseating names like "pupae". They harvested this grossness and used 2 different protein extraction techniques. Im just a little unclear on this, but it appears that the early embryonic stage organisms need to have the proteins extracted in a different way. Dechlorination is involved. The proteins were separated in the first dimension with SDS-PAGE and peptides digested out for 280 min nanoLC runs into a quadrupole Orbitrap (plus) system running a standard Top15 method. Ready for a new PTM to worry about Cysteine glycosylation is all over the place in human cells! Fun fact: Did you know that O-GlcNAc modified proteins were discovered in Baltimore over 30 years ago? See, theres more to my home town than fantastic infrastructure and friendly people! Glycoproteomics is kind of exploding right now -- the enrichments are better, the separations are better, and the mass specs are ridiculously better, and the software has almost caught up....and I wonder if this great new paper at ACS is just the tip of the iceberg.... A whole new class of glycopeptides right under our noses! The evidence looks pretty clear cut to me -- and first analysis from this group suggests that it isnt even rare.
